Usually, you use it for a situation that is more serious than the supermarket - "Kyrene_Hua, I know you are busy, but could you do me a favor and call our boss and tell him/her that we need an answer now for our project."

Second (lesson common - sarcasm): If someone is angry with you or frustrated about a situation with you. For example if you said, "Hey, WiseRichard, would you do me a favor and stop telling those bad jokes of yours?" (I had better do that if I want you to call our boss for me).

In a conversation, the tone of voice will tell you which one it is (polite or sarcasm). In writing, the context will tell you which one it is.
