Usually the premiere refers to a movie or television show being presented for the first time. For example, the new Disney film will premiere next month in Hollywood. A debut could also include a new song being performed, a new play opening on Broadway or even a young woman being formally introduced to "society". They are generally synonyms, but in common usage I would say that premiere typically is limited to "on screen" firsts.

They can be used interchangeably. But depending on the sentence, one may be more natural than the other. There are other contexts where only one really fits. For example, 'debut' tends to apply to the first appearance of an athlete in a particular competition or event, whereas you tend to talk about film 'premieres' instead. Going back to the words' French origins, début comes from 'start' and premier/première means 'first', but of course that doesn't prevent them from becoming entirely synonymous in many areas in English.
