Doesn't is the third person negative of the verb do.
Ain't is slang for isn't which is the the third person vegetative of the verb is.

Examples:
He doesn't want to go to bed.
I ain't doing anything in the afternoon.
I find that ain't is most commonly used in the southern US (although what do I know, I'm not even from there lol) and you definitely don't use ain't when writing, only speaking.
