The second sentence suggests that the comedian is forcing the people to laugh. So think of it like this, in the first sentence, the audience is laughing because they think the comedian’s joke is funny. In the second sentence, the comedian says a joke, but no one is laughing and the comedian tells everybody something like, “come on guys, laugh with me!”

"Had the people laughing" means that he was funny enough to cause the people to laugh. Saying that about a comedian is a good thing.
"His stand-up was so good that he had the whole crowd laughing together."
"Had the people laugh" sounds like the comedian was asking for people to laugh, kind of like you'd do by holding up a big sign that says, "Please Clap Now". It's a bit of a weird thing to say about a comedian.
"The guest comedian was pretty bad, but the stage director had the studio audience laugh anyways so that it would look better on the broadcast."
