Yes, it's quite common to hear 'to work on someone' used this way.
You tend to hear it more often in American English rather than British English, but it would be understood by all.

Here, it means to ask someone lots of questions etc in order to find out something that they maybe don't want to tell you.
It's a more informal way of using WORK On and doesn't follow the usual meaning of 'to practise something'.
