most people don't say "ain't" because it is considered to be non-proper english so we mostly use "isn't" but there's no difference in the meaning of both of them.

Ain't and isn't mean the same thing but isn't is the proper way to go rather than ain't. For example: "that isn't my key" sounds better than "that ain't my key"
