We would not say "I asked a question of you" very often, but it has the same meaning as, "I asked you a question." (This question is for you to answer.)
However, "I asked a question about you." means there was a question that was asked, and you were the topic.
For example, someone could ask a question to your boss, and it was about you. They could have asked, "Have you seen HELLOJYP today? He's not here yet."
This question is "about you".
