That's more of an expression. If something does happen again, you can say 'oh no, not again' or just 'not again' as a negative reaction. You can also use it if you think something bad is going to happen again too and you're talking about that thing, I hope that made sense haha

They both pretty much have the same meaning. Yet again gives the impression of the action having been repeated many times over, while again implies that it has been done before.
Example:
"She ate my food again."
"He messed the room up yet again."

'Yet again' is usually used in a negative context, when something keeps happening or someone keeps doing something that you want them to stop doing.
