ain't is actually an incorrect word. "is not" or "isn't" would be the correct words to use. however an example might be. "He ain't here." as in "he is not here."

Do not use "ain't" because it is not grammatically correct. But here is an example sentence:
"They ain't from America"

"Ain't" is not a real word. So you should not use it.
There are some people that use it like it is a word but it is not correct because it does not stand for anything. Here are some examples of contractions,
Are not "aren't"
I'm "I am"
Haven't "have not"
Don't "do not"
Didn't "did not"
Ain't is not a real contraction because you can not split it into two words.
