"Who do you want to speak with?" Is how most people would say it, although it is grammatically incorrect. Most people in casual situations don't say whom anymore.
"Whom do you want to speak with?" Or "With whom do you want to speak?" Are the two grammatically correct ways to say it. However, those two sentences would only be seen in formal writing or speeches

Who you want to speak with = any person you chose you talk to.
Whom do you want to speak with = certain people you want to talk to/Be friends with.

First sentence is grammatically correct although a little formal. Imagine a teacher asking a student who wants to file a complaint: Who do you want to speak with? This means both parties are engaging in a conversation.
Who do you want to speak to? is also correct but that directly means only one person is talking and the other is listening.
So in casual conversation, you might want to use, who do you want to talk to? I know 말하다 means "to speak with" and 이야기하다 is more like "to talk to" so that may cause confusion for you when translating to English.
The second sentence is grammatically wrong, it should be, To whom do you want to speak with?
The only difference is the formality. The second sentence is more formal but they mean essentially the same thing. Casual conversations prefer the word "talk to" instead of "speak with."
