There is a difference. The first sentence is grammatically wrong.
Also, "preposition" should be plural since you are referring to it in a general sense. (i.e. "You don't need to use prepositions.")
If you are referring to a specific preposition, then perhaps you could phrase it as: "You don't need to use a preposition here / in this sentence."

@ErialC The first sentence was from a video lesson about how to use or don't use prepositions. And the teacher is native speaker.
The whole sentence sounds as: Can you meet me ON tomorrow? I hope you sad it's wrong, because we need no preposition before the word tomorrow.

@sanyachel: Technically, "we need no preposition" is wrong, though it is a common colloquial speech pattern (i.e. slang).
For it to be grammatically correct, it should be:
"We don't need a preposition..."
or
"We don't need any prepositions..."
or
"We need not use a preposition..."
